fa ارزیابی فنوتیپی، تجزیه ارتباطی و حاشیه‌نویسی ژن‌های مرتبط با پژمردگی برگ گندم نان (.Triticum aestivum L) در مرحله گیاهچه‌ای در شرایط تنش خشکی Phenotyping, association analysis and annotation of genes related to leaf wilting of bread wheat (Triticum aestivum L.) at the seedling stage under drought stress conditions تخصصي Special پژوهشي Scientific & Research غربالگری سریع ژرم‌پلاسم گیاهی در مراحل اولیه رشد و شناسایی مبنای ژنتیکی شاخص پژمردگی برگ گندم در مرحله گیاهچهای برای برنامههای اصلاحی گندم ضروری است. در تحقیق حاضر، تعداد 290 ژنوتیپ گندم نان ایرانی شامل 90 رقم زراعی و 200 توده بومی در سال 1400 در گلخانه تحقیقاتی دانشکده کشاورزی دانشگاه ارومیه در شرایط تنش خشکی در مرحله گیاهچهای از نظر شاخص پژمردگی برگ مورد ارزیابی قرار گرفتند. نتایج نشان داد که بین ژنوتیپهای مورد مطالعه به لحاظ میزان پژمردگی برگ تنوع وجود داشت و تودههای بومی بهطور معنیداری، (حدود 12 درصد) از نمره پژمردگی بیشتری نسبت به ارقام زراعی برخوردار بودند که این موضوع میتواند نتیجه مستقیم یا غیرمستقیم گزینش باشد. از نظر شاخص پژمردگی برگ حدود نه درصد از ارقام زراعی و سه درصد از تودههای بومی در گروه متحمل به تنش خشکی قرار گرفتند. تجزیه ارتباطی با دو روش GLM و MLM تقریبا نتایج یکسانی داشت. کروموزمهای 3B، 2A، 7B، 1A، 3A، 4B، 5B، 6A و 6B بهترتیب بیشترین تعداد ارتباط نشانگر با شاخص پژمردگی برگ را در کل جمعیت داشتند که تنوع فنوتیپی صفت را از 6 الی 20 درصد توجیه میکردند. حاشیهنویسی نشانگرهای معنیدار، تاییدکننده نتایج تجزیه GWAS بود و نشان داده شد که تعداد پنج نشانگر روی کروموزومهای 1A، 2A، 3A و 7B با ژن‌هایی که در تحمل گیاه گندم به تنش خشکی نقش دارند، همردیفی داشتند. جزء جدایی ناپذیر غشا بودن، فسفوریلاسیون پروتئینها، فعالیت آنزیم‌های متیل ترانسفراز و اﺗﺼﺎل ﻳﻮن روی (Zn2+) بخشی از کارکردهای مولکولی و فرآیندهای زیستی ژنهای یاد شده بودند. یافتههای تحقیق حاضر، بینشی عمیق در رابطه با پژمردگی برگ گندمهای نان ایرانی در شرایط تنش خشکی ارائه می‌دهد که میتوان از آن در برنامههای گزینشی آتی گندم نان استفاده کرد.   Rapid screening of plant germplasm in the early stages of growth and determining the genetic basis of wheat leaf wilting index at the seedling stage is necessary for wheat breeding programs. In the present research, leaf wilting index for 290 Iranian bread wheat genotypes, including; 90 cultivars and 200 landraces were studied under drought stress conditions at the seedling stage in 2021 in research greenhouse of faculty agriculture of Urmia University, Urmia, Iran. The results showed that there was variation between the studied genotypes for leaf wilting index, and the landraces had significantly 12% more wilting scores than cultivars. This can be attributed to direct or indirect selection by breeders. The leaf wilting index of about 9% of cultivars and 3% of landraces were identified in drought stress tolerant group. GWAS had nearly the same results as general lineare model and MLM methods. Chromosomes 3B, 2A, 7B, 1A, 3A, 4B, 5B, 6A and 6B, respectively, had the highest number of marker-trait associations in the entire population which explained the phenotypic variation of the trait from 6 to 20%. Annotation of significant markers confirmed the results of GWAS analysis and showed that five markers on chromosomes 1A, 2A, 3A and 7B were aligned with different genes involved in drought stress tolerance. An integral component of membrane, protein phosphorylation, methyltransferase activity and zinc ion binding were part of the molecular functions and biological processes of the identified genes.
